Genetic and therapeutic targeting of properdin in mice prevents complement-mediated tissue injury.
The alternative pathway (AP) of complement activation is constitutively active and must be regulated by host proteins to prevent autologous tissue injury. Dysfunction of AP regulatory proteins has been linked to several human inflammatory disorders. Properdin is a positive regulator of AP complement activation that has been shown to extend the half-life of cell surface–bound C3 convertase C3bBb...
متن کاملNative properdin binds to Chlamydia pneumoniae and promotes complement activation.
Activation of complement represents one means of natural resistance to infection from a wide variety of potential pathogens. Recently, properdin, a positive regulator of the alternative pathway of complement, has been shown to bind to surfaces and promote complement activation. Here we studied whether properdin-mediated complement activation occurs on the surface of Chlamydia pneumoniae, an obl...
متن کاملComplement activation by tubular cells is mediated by properdin binding.
Activation of filtered complement products on the brush border of the tubular epithelium is thought to be a key factor underlying proteinuria-induced tubulointerstitial injury. However, the mechanism of tubular complement activation is still unclear. Recent studies on mechanisms of complement activation indicate a key role for properdin in the initiation of an alternative pathway. We hypothesiz...
متن کاملThe complement protein properdin binds apoptotic T cells and promotes complement activation and phagocytosis.
Apoptotic cells must be rapidly eliminated to avoid harmful inflammatory and autoimmune reactions. Innate immunity is designed/poised to identify dying cells by their unique surface-associated molecular patterns. Here we demonstrate for the first time, to our knowledge, that the human complement protein properdin binds to early apoptotic T cells and initiates complement activation, leading to C...
متن کاملQuantitative Studies on the Properdin-complement System
The reaction of human, bovine, and pig properdin with excess zymosan exhibits an induction period followed by a rapidly accelerating formation of the properdin-zymosan (PZ) complex. Two methods for determining the quantity of PZ complex are given, one using human serum lacking properdin, the other using a fraction of pig serum containing complement components, C'1, C'3 and C'4.
